Danocrine

CLASS and MECHANISM of Action

Generic "Danocrine" works best at flattening lesions and removing scales. This drug is a synthetic form of vitamin D3 that can be applied to the skin. Applying calcipotriol/calcipotriene once to twice a day controls the speed of turnover of skin cells. It is sometimes combined with topical corticosteroids to reduce irritation.
Suppresses pituitary output of follicle-stimulating hormone and luteinizing hormone that causes regression and atrophy of normal and ectopic endometrial tissue; decreases rate of growth of abnormal breast tissue; reduces attacks associated with hereditary angioedema by increasing levels of C4 component of complement.

Active Ingredients: vitamin D3.
Inactive Ingredients: lactose, maize starch, purified talc, magnesium stearate, titanium dioxide, gelatin.

What is Generic "Danocrine" PRESCRIBED For?

Generic "Danocrine" is indicated for use in the treatment of visually proven (e.g., laparoscopy) endometriosis where the required end-point of treatment is fertility, or for the control of symptoms when surgery is contraindicated or has been unsuccessful. Generic "Danocrine" is indicated for the short-term management of intractable primary menorrhagia.
Generic "Danocrine" is indicated for the prophylaxis of attacks of hereditary angioedema of a severe or life-threatening nature, in male and female patients.
Generic "Danocrine" is indicated for the short-term treatment of severe benign (fibrocystic) breast disease or mastalgia associated with severe symptomatic benign breast disease, in patients refractory to other treatments.

How Should I Take Generic "Danocrine"?

In women of reproductive age, therapy should begin during menstruation. A sensitive test capable of determining early pregnancy is recommended immediately prior to start of therapy. Additionally, a non-hormonal method of contraception should always be used during Generic "Danocrine" therapy.
Endometriosis : 200-800 mg Generic "Danocrine" daily in two to four divided doses. It is recommended that treatment be initiated with a dosage of 800 mg daily in 4 divided doses. In some patients, it may be possible to maintain improvement with a reduced dosage once a satisfactory response has been obtained. Treatment should continue uninterrupted for 3 to 6 months, but may be extended to 9 months if necessary.
Menorrhagia : A course of 200-400 mg Generic "Danocrine" daily in divided doses for up to 6 months. 200 mg is usually sufficient to reduce menstrual blood flow to acceptable limits.
Fibrocystic Breast Disease : The minimum effective dose should be used. 200 mg daily is an effective dose in the majority of patients. In some instances, 400 mg/day may be warranted.
Hereditary Angioedema : 200-600 mg Generic "Danocrine" daily in divided doses. Dosage should be kept as low as possible with adjustment to meet individual patient requirements. Consideration should be given to interrupting treatment after an attack-free period.

What are the CONTRAINDICATIONS for taking Generic "Danocrine"?

Generic "Danocrine" is contraindicated in patients with:
  • Undiagnosed abnormal genital bleeding.
  • Undiagnosed ovarian/uterine masses.
  • Past jaundice with oral contraceptives.
  • Markedly impaired liver, renal or cardiac function, including oedema.
  • Pelvic infection.
  • Neoplasia of primary or secondary sexual organs.
  • Hypertension WHO II or worse.
  • Known hypersensitivity to danazol.
  • Pregnancy.
  • Breast feeding.
  • Porphyria - Generic "Danocrine" can induce ALA synthetase activity and hence porphyrin metabolism.
  • Androgen-dependant tumour.
  • Active thrombosis, thromboembolic disease or history of such events.

What Happens if I MISS A DOSE?

Apply the missed dose as soon as you remember. However, if it is almost time for your next dose, skip the dose you missed and apply only your next regularly scheduled dose.

What happens if I OVERDOSE?

You suspect an overdose, or if Generic "Danocrine" has been ingested, call an emergency room or poison control center near you.
Symptoms of an overdose of Generic "Danocrine" include weakness, fatigue, drowsiness, dizziness, headache, decreased appetite, nausea, vomiting, and high levels of calcium in the blood.

What are the possible SIDE EFFECTS of Generic "Danocrine"?

In general, the side effects associated with Generic "Danocrine" therapy are attributable to the pharmacological activity of the drug; these effects may reflect Generic "Danocrine" 's weak androgenic and anabolic activity and/or the gonadal suppression which results from therapy.
Serious side effects are very rare. Stop using Generic "Danocrine" if you experience an allergic reaction (shortness of breath; closing of your throat; swelling of your lips, face, or tongue; or hives).
You may experience some irritation of the skin, such as burning, itching, redness, swelling, dryness, or peeling while you are using Generic "Danocrine". These side effects sometimes lessen with continued treatment. Notify your doctor if you experience these side effects.
If you notice a change in your skin color, contact your doctor.
Side effects other than those listed here may also occur.
Talk to your doctor about any side effect that seems unusual or that is especially bothersome.

What other drugs can INTERACT with Generic "Danocrine"?

Carbamazepine: Generic "Danocrine" may increase carbamazepine concentrations, requiring a decreased dosage of carbamazepine; monitor
Cyclosporine: Generic "Danocrine" may increase cyclosporine concentrations leading to increased toxicity; dosage of cyclosporine may need decreased; monitor
Hormonal contraceptives: Generic "Danocrine" also inhibits ovulation, possibly by acting at (and competing for) the same hormonal receptors; nonhormonal birth control methods are recommended during therapy.
Hypoglycemic agents: Generic "Danocrine" effects glucose metabolism, concurrent use may lead to hypoglycemia; monitor
Tacrolimus: Generic "Danocrine" may increase tacrolimus concentrations leading to increased toxicity; monitor
Warfarin: Increased anticoagulant effect; avoid concomitant use if possible
