Sinemet

CLASS and MECHANISM of Action

Levodopa is converted to a chemical called dopamine (DOE pa meen) in the brain. Symptoms of Parkinson's disease may be caused by low levels dopamine in the brain. Levodopa is turned into dopamine in the body.
Carbidopa helps prevent the breakdown of levodopa before it can reach the brain and take effect.
The combination of carbidopa and levodopa is used to treat Parkinson's disease and Dopa-Responsive Dystonia (DRD). Carbidopa and levodopa is also used to treat Parkinson symptoms caused by carbon monoxide poisoning or manganese intoxication.

What is Generic Sinemet PRESCRIBED For?

Generic Sinemet is indicated for the treatment of Parkinson's disease and syndrome. It is useful in relieving many of the symptoms of parkinsonism, particularly rigidity and bradykinesia. Generic Sinemet is helpful in the management of tremor, dysphagia, sialorrhoea, and postural instability associated with Parkinson's disease and syndrome.
When therapeutic response to levodopa alone is irregular, and signs and symptoms of Parkinson's disease are not evenly controlled throughout the day, substitution of Generic Sinemet usually is effective in reducing fluctuations in response.
By reducing certain adverse reactions produced by levodopa alone, Generic Sinemet permits more patients to obtain adequate relief of the symptoms of Parkinson's disease. Generic Sinemet is also used for patients with Parkinsonism who are taking vitamin preparations that contain pyridoxine hydrochloride (Vitamin B6).

How Should I Take Generic Sinemet?

The optimum daily dosage of Generic Sinemet must be determined by careful titration in each patient. Tablets of the two ratios may be given separately or combined as needed to provide the optimum dosage. Each tablet of Generic Sinemet is designed to divide in half with minimal pressure. Keep Generic Sinemet in the container it came in, tightly closed, and out of reach of children. Store it at room temperature and away from excess heat and moisture (not in the bathroom). Throw away any medication that is outdated or no longer needed. Talk to your pharmacist about the proper disposal of your medication.

What are the CONTRAINDICARIONS for taking Generic Sinemet?

Nonselective monoamine oxidase (MAO) inhibitors are contraindicated for use with Generic Sinemet. These inhibitors must be discontinued at least two weeks prior to initiating therapy with Generic Sinemet. Generic Sinemet may be administered concomitantly with the manufacturer's recommended dose of a MAO inhibitor with selectivity for MAO type B (e.g., selegiline HCl)
Generic Sinemet is contraindicated in patients with known hypersensitivity to any component of this medication, and in patients with narrow angle glaucoma. Since levodopa may activate a malignant melanoma, Generic Sinemet should not be used in patients with suspicious undiagnosed skin lesions or a history of melanoma.

What Happens if I MISS A DOSE?

Take the missed dose of Generic Sinemet as soon as you remember it. However, if it is almost time for the next dose, skip the missed dose and continue your regular dosing schedule. Do not take a double dose to make up for a missed one.

What happens if I OVERDOSE?

Seek emergency medical attention if you think you have used too much of Generic Sinemet.
Overdose symptoms may include uneven heart rate, muscle spasms, and severe nausea or vomiting.

What are the possible SIDE EFFECTS of Generic Sinemet?

    Generic Sinemet may cause side effects. Tell your doctor if any of these symptoms are severe or do not go away:
  • nausea
  • dizziness
  • vomiting
  • diarrhea
  • loss of appetite
  • constipation
  • dry mouth
  • change in sense of taste
  • forgetfulness or confusion
  • nervousness
  • headaches
  • weakness
  • nightmares
  • difficulty falling asleep or staying asleep
  • increased sweating
  • drowsiness
    Some side effects can be serious. If you experience any of the following symptoms, call your doctor immediately:
  • unusual or uncontrolled movements of the mouth, tongue, face, head, neck, arms, and legs
  • depression
  • fast, irregular, or pounding heartbeat
  • hallucinating (seeing things or hearing voices that do not exist)
  • thoughts of death or killing oneself
  • swelling of the face, throat, tongue, lips, eyes, hands, feet, ankles, or lower legs
  • difficulty swallowing or breathing
  • hoarseness
  • hives
  • bloody vomit
  • vomiting material that looks like coffee grounds
  • red blood in stools
  • black and tarry stools.

What other drugs can INTERACT with Generic Sinemet?

    Before taking Generic Sinemet, tell your doctor if you are taking any of the following medicines:
  • phenytoin (Dilantin);
  • isoniazid (Nydrazid);
  • metoclopramide (Reglan);
  • papaverine (Pavabid, Papacon, Pavagen, Pavacot);
  • blood pressure medication;
  • antidepressants such as amitriptyline (Elavil, Etrafon), amoxapine (Ascendin), clomipramine (Anafranil), desipramine (Norpramin), doxepin (Sinequan), imipramine (Janimine, Tofranil), nortriptyline (Pamelor), protriptyline (Vivactil), or trimipramine (Surmontil); or
  • phenothiazines such as chlorpromazine (Thorazine), fluphenazine (Prolixin), mesoridazine (Serentil), perphenazine (Trilafon), prochlorperazine (Compazine), thioridazine (Mellaril), or trifluperazine (Stelazine).
This list is not complete and there may be other drugs that can interact with Generic Sinemet. Tell your doctor about all your prescription and over-the-counter medications, vitamins, minerals, herbal products, and drugs prescribed by other doctors. Do not start a new medication without telling your doctor

Additional Information

Keep all appointments with your doctor and the laboratory. Before having any laboratory test, tell your doctor and the laboratory personnel that you are taking Generic Sinemet.
As your condition improves and it is easier for you to move, be careful not to overdo physical activities. Increase your activity gradually to avoid falls and injuries.
Generic Sinemet can cause false results in urine tests for sugar (Clinistix, Clinitest, and TesTape) and ketones (Acetest, Ketostix, and Labstix). Diabetic patients should use TesTape to test urine for glucose (sugar); better results can be obtained by holding the tape vertically, inserting the lower portion of the tape into the urine sample, and reading the color at the top of the damp area.
